The objective of this research is to elucidate the importance of non-covalent bonds in proteins on tolerances towards extreme environments. A 2Fe-2S feredoxin from triangular disc-shaped extremely halophilic archaeon Haloarcula japonica was investigated. It seemed that the additional amino terminal region of H. japonica Fd was importantfor halotolerancy. The thermostability of 3-isopropylmalate dehydrogenase of moderate facultative thermophile Bacillus coagulans was improved by amino acid substitutions at the subunit-subunit interface and/or a loop region.Furthermore, the function of chitin-binding domain of a multi-domain chitinase from alkaliphilic Bacillus sp. J813 was elucidated.
