Research Abstract

ロドプシンが光を受容するとG蛋白質(トランスデューシン)を活性化するようになるが、この過程で起こる蛋白質部分の構造変化を原子レベルで解析することを試みた。そのため、ロドプシンが活性状態に変化する過程で生成する4つの反応中間体について、X線回折データの収集を行い、それらのデータの解析を行った。また、初期中間体では蛋白質部分の大きな構造変化は起こらないので、データの精密化の基礎となるウシロドプシンの3次元結晶の品質を上げる努力を行った。昨年度我々はウシロドプシンの結晶化条件の再検討・基底状態構造の精密化を行った結果、2.6Åの分解能でロドプシンの立体構造を解析し、7つの水分子の同定に成功した。本年度は結晶化条件のさらなる検討を行い、分解能が向上した回折像の獲得に成功した。さらに種々の結晶化条件を検討して、この分解能を上げる努力を行った。
また、培養細胞系でウシロドプシンを発現させ、抗体カラムを用いた精製法を適用することにより、結晶化に適した試料を調製することに成功した。また、その結晶化を試みた結果、結晶化する条件を見つけることができた。また、構成的活性化を示す(Constitutive Activeな)ロドプシン変異体について、その培養細胞系での発現・精製にも成功し、結晶化の条件を探索した。さらに、結晶化のための大量の試料を調製するために、ロドプシン変異体遺伝子を導入したStable Cell Lineを作製した。
