Research Abstract

生理的なヘム分解酵素であるヘムオキシゲナーゼ(HO)は、1960年代の半ばに米国のSchmidらによって発見された。以来国内外の数多くの研究者により精力的に研究されてきた。我々は、構造生物学観点からこの酵素の反応機構を追求し、本申請課題において次のような成果をえた。
1.ラットHO-1のC末端22アミノ酸残基(膜結合部位)を除いた可溶性酵素(Met1-Pro267)の発現に成功し、10Lの培養液から170mgの精製標品を得るシステムを確立した(Kurume med. J.43,313,1996)。
2.酸素・電子滴定装置を用いて、ヒドロキシヘムからベルドヘムへの転換には、シトクロムP450還元酵素からのe^-の供給は必要とせず、分子状酸素のみで起こることを発表した(J. Biol. Chem. 274, 18196, 1999)。
3.ヘム-HO結合体の高分解能の回折像(2.4Å)を得た(Acta. Cryst. D54, 1017, 1998)。
4.ラットHO-1のセレノメチオニン変異酵素を用いて位相の決定し,ラットHO-1の結晶構造を決定した(FEBS Lett. 471, 61, 2000)。
5.4種のベルドヘム異性体の有機合成に成功した(J. Inorg. Biochem. 82, 113, 2000)。
6.ヒドロキシヘムからベルドヘムへの転換には、外部からのe^-の供給は必要とせず、O_2のみで起こることを再確認し,電子要求性についての研究者間の不一致は,結合体の調製方法と用いる還元系(ジチオナイトなど)の違いによることを明らかにした(Eur. J. Biochem. 269, 5231-5239, 2002)。
7.ラットHO-1について,アポ型(Biochemistry41,7293-7300, 2002)およびヘム-N_3結合型(J. Biol. Chem.277, 45086-45090, 2002)の結晶構造を明らかにした。
