Research Abstract

近年の研究で、末梢から脊髄レベルでの痛覚修飾の分子基盤は明らかになってきたが、視床より上位中枢では、ほとんど明らかになっていない。一方、三量体G蛋白のGq/11結合型受容体に共役する、phospholipase Cβ4(PLCβ4)は、視床に優位に発現していることから、この分子が、体性感覚情報、とりわけ体性痛覚の情報処理に寄与するか否かを調べるため、PLCβ4の遺伝子欠損マウスを用いて、疼痛関連行動の解析を行った。ノックアウトマウスでは、炎症疼痛モデルのホルマリンテストを行ったところ、第一相の疼痛行動には差はないが、第二相の疼痛反応時間は野生体に比べて29.9土3.3%まで減少上していた。さらに我々は、マウスでの脳室内投与法、視床内投与法を確立し、野生体にPLCまたその上流にグルタミン酸受容体1型(mGluR1)の阻害剤、拮抗薬を投与すると同様にホルマリンテストの第二相が減弱した。これらの結果から、視床mGluR1-PLCβ4のカスケードが炎症性疼痛の修飾に関わっていることが明らかになった。さらに、マウスの視床から細胞外記録を行い、疼痛時にPLCβが細胞の発火を調整する事により、痛みをコードしていることが明らかになった。
