Research Abstract

本研究では、パーキンソン病の病態生理に関して近年もっとも注目されてきた、「パーキンソン病の発症過程において、大脳基底核のグルタミン酸作動性神経伝達が亢進する」という報告に基づき、まずパーキンソン病の際に大脳基底核の代謝調節型グルタミン酸受容体(mGluRs)の発現様式がどのように変化するかを免疫組織化学的に解析した。その結果、MPTP投与により実験的に作製したパーキンソン病のモデルサルでは、mGluR1αの発現が淡蒼球と黒質において特異的に低下していることを明らかにした。次に、覚醒下の健常なサルで、mGluR1やgroup I mGluRsに対する選択的な薬物を淡蒼球に局所注入し、淡蒼球ニューロンの自発発火活動がどのように変化するかを電気生理学的に解析した。その結果、淡蒼球ニューロンの発火頻度は、group I mGluRsの作動薬であるDHPGにより著明に増加し、group I mGluRsの桔抗薬であるAIDAやmGluR1の拮抗薬であるLY367385により有意に減少した。以上のデータは、視床下核由来と思われるグルタミン酸がmGluR1αを介して淡蒼球ニューロンを興奮させることを示唆しており、パーキンソン病における淡蒼球や黒質のmGluR1αの発現低下は、視床下核ニューロンの活動性が増大することによる淡蒼球ニューロンの過剰興奮を補償するための変化であると考えられる
