Research Abstract

アレルギー性疾患の発症にはIL-4とIL-5を産生するCD4陽性T細胞(Th2細胞)の選択的活性化が深く関与している。本研究において我々は、IL-2をはじめ多くのサイトカインにより活性化されるSignal transducers and activators of transcription 5a (Stat5a)の遺伝子欠損マウスでは、抗原特異的刺激によるTh2細胞分化が選択的に障害されていることを明らかにした(Blood 97:2358,2001)。そしてStat5aにより発現誘導される事が知られでいる分子の発現についてスクリーニングを行った結果、Stat5a欠損マウスでは、CD25陽性免疫制御性T細胞が減少し、この減少がStat5a欠損マウスにおけるTh2細胞分化の障害に関与していることを明らかにした(Am. J. Respir. Crit. Care Med.164:680,2001)。加えてStat5a/Stat6ダブル欠損マウスを用いた解析により、Stat5a欠損マウスにおけるTh2細胞分化の障害はStat6非存在下でも認められることを明らかにし、Th2細胞分化におけるStat5a依存性'Stat6非依存性経路の存在を明らかにした(論文投稿中)。現在、CD25陽性免疫制御性T細胞によるT細胞分化制御の分子メカニズムの解析、Stat5a依存性Stat6非依存性経路の分子メカニズムの解析とともに、未知の経路の存在を考えStat5aにより発現誘導され、かつTh2細胞の分化を制御する分子をレトロウイルス発現クローニング法を用いて検索中である。
