Research Abstract

記憶B細胞産生と維持におけるFasおよびp2lrasの関与をそれぞれFas不全マウスlprマウスおよび我々が作製した優勢抑制型Ha-ras発現トランスジェニックマウスを用いて解析した。その結果、Fasを介したシグナルは、胚中心における低親和性B細胞の排除を促進し、その結果高親和性B細胞の選択に間接的に寄与することが明らかに去れた。さらに、Fasを介したシグナルは体細胞変異を高度に蓄積する胚中心B細胞の記憶B細胞への分化を抑制することが明らかにされた。記憶B細胞は免疫後初期に産生され、その後一定の数で長期に維持されるが、この維持には、Fasを介したシグナルとそれに拮抗する未知のシグナルが関与する可能性を明らかにした。
胚中心では高親和性を示すB細胞が選択され、記憶B細胞や長期の寿命をもつ抗体産生細胞へ分化すると考えられている。本研究において、p21rasは記憶B細胞の産生に関与しないが、高親和性を示す記憶B細胞の選択に必要であることを明らかにした。しかし、高親和性を示す胚中心B細胞の選択や、長期の寿命をもつ抗体産生細胞の選択はp21ras非依存的に誘導される可能性が示唆された。これらの結果は、胚中心での高親和性を有する記憶B細胞と抗体産生細胞の選択が異なった機構により制御される可能性を示唆するとともに、高親和性で長期の寿命をもつ抗体産生細胞は記憶B細胞とは独立した系として存在する可能性を支持する。
