Research Abstract

海馬CA1領域におけるLTP誘導には,Ca^<2+>/カルモデュリン依存性プロテインキナーゼII(CaMキナーゼII)活性化反応が重要な役割を演じている。
本研究では,海馬CA1領域,LTP誘導,維持におけるCaMキナーゼII, CaMキナーゼIV, mitogenactivated protein kinase(MAPキナーゼ)活性化反応,転写因子活性化反応,蛋白質合成刺激反応を調べるために,LTP誘導後の海馬切片を用い,酵素活性,遺伝子発現を経時的変化の中で検索した。すなわち,1)CaMキナーゼII, IV, MAPキナーゼ活性化反応が観察され,経時的変化は相違した。CaMキナーゼII活性化反応は持続し,CaMキナーゼIV, MAPキナーゼは誘導後3分でピークを示して10分では元のレベルに戻った。2)CREB燐酸化反応の経時的変化とCaMキナーゼIV阻害薬,MAPキナーゼ阻害薬のCREB燐酸化反応に対する効果を経時的変化の中で調べた。3)brain-derived neurotrophic factor(BDNF)はそれ自身,単独投与ではLTP誘導能を持たなかったが,短時間テタヌス刺激(単独ではLTP誘導能を持たない)を併用すると,LTPが誘導された。それぞれの阻害薬を用いることで,CaMキナーゼII, IV, MAPキナーゼ活性化反応とLTP誘導,維持との相関性を調べ,その機構を検索した。4)CREB燐酸化反応を調べ,LTP維持において,CaMキナーゼIVとMAPキナーゼが遺伝子発現に関与することを明らかにした。
以上の結果は,長期増強誘導機構には,CaMキナーゼIIが関与し,一方,維持機構にはCaMキナーゼIV, MAPキナーゼが関わって遺伝子発現を刺激していることを示唆している。
