Research Abstract

出芽酵母にはTOR1/2の2種類の遺伝子が存在し,両者は酵母の生育に必須である.もし,Torの活性制御因子に変異が入れば,その変異体は致死になるであろう.しかし,その欠損はTOR遺伝子過剰発現により抑制される可能性が強い.そこで,TOR2遺伝子のガラクトースによる過剰発現系プラスミド(pGAL[TOR2])を構築し,それを導入した酵母細胞を変異源処理して,pGAL[TOR2]を生育に必須とする変異体のスクリーニングを行った.単離した3変異株はすべて,温度感受性を示し,TOR2過剰発現により抑制されることが判った.次に上で得られた変異体より遺伝子の単離を行うためのゲノムライブラリーを作成した.
