Research Abstract

アニオン性ポルフィリンであるテトラカルボキシフェニルポルフィリンに対する抗体にポルフィリンを取り込ませ、この複合体に電子受容体であるメチルビオローゲンとヒドロゲナーゼを加えて、可視光を照射した。抗体中のポルフィリンから電子受容体に移動した電子と触媒である還元酵素ヒドロゲナーゼを用いて、水から水素を発生させることに成功した。さらに本年度は抗体を用いて樹状超分子を構築した。抗体IgMが結合する抗原を抗体IgGに化学修飾し、IgMと抗原修飾IgGを混合することによって樹状抗体超分子を得た。原子間力顕微鏡により抗体デンドリマーの構造を直接観察することに成功した。酵素標識抗体測定法(ELISA)および表面プラズモン共鳴(SPR)を検出原理とするバイオセンサーにおいて本抗体デンドリマーを用いると各測定法におけるシグナル強度は抗原存在下において増大した。本抗体デンドリマーの基質特異性は極めて高く、優れたバイオセンシング材料になることがわかった。さらに抗体を認識するタンパク質を核とする抗体超分子、および金ナノ粒子をコアとしてその表面に抗体を集積した無機-生体高分子ハイブリッドを合成した。抗原を修飾したセンサーチップに抗体IgG、抗体-プロテインA超分子、抗体-金ナノ粒子の3種の溶液(抗体濃度一定)をそれぞれ添加したときのSPR応答シグナル強度を比較した。金-抗体ハイブリッドを用いたセンシングシステムでは単一の抗体添加系の14倍のシグナル強度が得られた。抗体の集積および金粒子とのハイブリッド化により応答シグナル強度を増幅させることができた。
