Research Abstract

マウス小腸に発現するAQP分子種を解析したところ、AQP1,3,4,5,7,8,9が発現しており、AQP0,2,6は発現していなかった。AQP9についてはRT-PCRおよび以下のin situハイブリダイゼーション(ISH)で小腸におけるその発現が初めて明らかとなった。
小腸の細胞機能におけるAQPの役割を考察するために、各細胞種に発現するAQP分子種を調べた。小腸粘膜上皮において、パネート細胞にはAQP1,3,4が、吸収上皮細胞にはAQP1,3が発現していると推測された。杯細胞および内分泌細胞に発現するAQPの詳細は単一分子種の発現分布からは同定できなかった。そこで、マーカー分子を用いて解析を行った結果、内分泌細胞に関しては発現分子種の同定には到っていないが、小腸上皮においてはいずれの細胞種も複数種のAQPが発現していることが明らかとなった。また、杯細胞にはAQP9を発現している細胞と発現していない(もしくは発現量が非常に少ない)細胞が存在することが明らかとなった。
AQP9を発現している杯細胞に注目し、消化管におけるAQPの機能を解析した。大腸の杯細胞では強いAQP9の発現が観察されたが、胃の粘液分泌細胞である表層粘液細胞ではAQP9の発現は観察されなかったことから、AQP9は小腸および大腸の粘液分泌細胞種である杯細胞に特異的な機能を担っていると推定された。
ISHおよび免疫染色の結果、AQP9は杯細胞の一部に発現していると考えられたため、杯細胞での発現が知られている10種のマーカー分子の発現様式を解析し、AQP9がどのような杯細胞に発現しているのかを解析した。その結果、AQP9の発現は分化段階には依存しないことが示された。このことと、AQP9が水以外にも様々な小分子を透過させる性質を持つことを合わせて考えると、AQP9は粘液組成を多様化させることに関与していると推察された。
