Research Abstract

耳下腺に局在するAquaporin-5(AQP5)は、Acetylcholine(ACh)でムスカリン受容体を刺激すると管腔膜で急激に増量され一過性に唾液分泌を促進する。新規薬物SNI-2011は管腔膜でゆっくりとAQP5を増量し持続的に唾液分泌を促進する。この機序の解明と老化や糖尿病による口腔乾燥症の発症機序の解明を行った。1)共焦点顕微鏡と電子顕微鏡による解析から、小葉管導管にてSNI-2011刺激によりAQP5はlipid raftのマーカー・GM1やFlotillin-2と共に細胞質より管腔膜へ移動した。老齢ラットや糖尿病ラットにおいて、このAQP5の細胞内移動が阻害されていた。特に、老齢ラットではSNI-2011刺激によりAQP5の基底膜への移動が認められた。2)Triton X-100(TX)を用いた生化学的解析から、AChやSNI-2011刺激によりAQP5はTX不溶性画分から可溶性画分へAChでは15秒、SNI-2011では10分で最大となる移行が認められた。即ち、刺激によりraftsからnon-raftsへ移動した。また、老齢ラットにおいては、無刺激時raftsへのAQP5の蓄積が顕著であったが、SNI-2011刺激によりraftsからnon-raftsへ移動が認められた。糖尿病ラットにおいては、raftsにおけるAQP5の量が低下すると共にraftsからnon-raftsへ移動が認められなかった。3)A23187で刺激してもSNI-2011刺激とほぼ同様の結果が共焦点顕微鏡や電子顕微鏡による解析、生化学的解析において得られた。以上のことから、無刺激時の耳下腺小葉管導管において、AQP5は細胞質に局在しており、ムスカリン受容体の刺激により細胞質のCa^<2+>濃度が上昇するとraftsとともに管腔膜へ移動し、raftsからnon-raftsへ移行して機能すると考えられる。加齢とともにAQP5はraftsに蓄積され、ムスカリン受容体の刺激により管腔朦への移動が減り、基底膜への移動が増量して、糖尿病では、lipid raftsにおけるAQP5の減量と管腔膜への移動の低下により口腔乾燥症が発症するものと考えられた。
