Research Abstract

当該するテーマについて、in vitroでアクアポリン-2(AQP-2)5'上流領域の遺伝子転写調節機構の解明を継続して進めた。高浸透圧に加えて、低浸透圧の変化に呼応するAQP-2 promoter遺伝子の浸透圧反応領域の特徴を検討した。マウスAQP-2遺伝子5'上流域(-9.5kb)をpGL-3 basic vectorのルシフェラーゼ(LUC)遺伝子上流に組み込んだコンストラクトを作製した。AQP-2 promoter遺伝子の種々の大きさのDNA fragmentsを組み込んだプラスミッドをMDCK細胞にトランスフェクトして実験に供した。これまでの研究から、高浸透圧に対するAQP-2転写調節を司る浸透圧反応領域は少なくとも2カ所存在することが示された。これは、-570〜-560bpに存在するtonicity-responsive enhancer (TonE)と、新たな反応部位である-6.1〜-4.3kb内にTonEとは独立して浸透圧反応を調節する部位である。とくに、AQP-2遺伝子転写調節には後者の新たな反応領域の介入が優位であることが示された。また、両者のシグナル伝達系は異なり、TonEがMAPキナーゼ系を、新しい部位はMAPキナーゼ以外の未知のシグナル伝達系によると考えられる。TonE結合蛋白を過剰発現させたMDCK細胞では、-1.1 AQP2に比べて-6.1 AQP2で高浸透圧によるLUC活性が著しく増強されることから、これらの2つの浸透圧反応領域はAQP-2遺伝子転写調節を互いに協調して促進する可能性も考えられる。
さらに、低浸透圧によるAQP-2 promoter遺伝子調節も検討した。LUC活性は低浸透圧(250,225mmol/kg)下でも増加し、これはMAPキナーゼ阻害薬で抑制された。しかし、この現象は細胞が膨化できないため低浸透圧が持続するためと考えられた。このため、AQP-2 promoterとAQP-2を共発現させたところ、低浸透圧による細胞の膨化が完了した後は、db-cAMPによるAQP-2 promoter活性は対照群より有意に減弱した。この結果は、in vivo実験で示したSIADHモデルラットにおけるAVP escape現象にみられるAQP-2発現減弱の分子機構につながる方向性が示唆された。
