Research Abstract

1.マウスの前後軸回転がWnt/β-cateninシグナルにより制御されていることを示した。Otx2変異マウスで見られる軸回転異常はβ-cateninシグナルの抑制不全に起因していることをβ-cateninの活性を観察すること等で明らかにした。更に、Otx2標的遺伝子であるDkk1の発現を詳細に解析することから、このWnt拮抗因子が臓側内胚葉の前方移動の誘因分子となっていることが示唆された。
2.硬骨魚類において哺乳動物の前方臓側内胚葉がどの組織に対応するのかを解明するため、フグOtx2遺伝子の前方臓側内胚葉特異的なエンハンサーがゼブラフィシュにおいてどのような発現活性を持つか解析した。結果、この制御領域は硬骨魚においては前方中内胚葉の発現を支配していた。
3.Otx2遺伝子の前方臓側内胚葉における発現調節領域を解析した結果、Fork headファミリーの転写因子の結合部位が発現に必須であることが明らかになった。
4.内在性のWntシグナル経路が実際に終脳の領域化に関わるかについて検討を行った。結果、Wntシグナル伝達に関わる因子のうち、シグナルを負に制御する因子、あるいは変異分子の強制発現実験によって、内在性のWntシグナルが終脳の初期の領域化に重要であるとの結果を得た。さらに、内在WntとしてWnt8b、Wnt1を候補因子と考え、siRNAによるノックダウン実験を行うための材料、実験条件等の準備を行った。
5.Ssdep1変異マウスを用いて脊索前板と頭部の形成機構を解析した。Ssdp1は頭部形成においてLim1、Ldb1と遺伝的相互作用を示し、Lim1-Ldb1-Ssdp1複合体が脊索前板を形成することが示唆された。Ssdp1変異胚では前頭部マーカーの消失以前に、頭部欠失を説明しうる細胞死・増殖の変化は見られず、脊索前板の主たる機能は、前頭部の遺伝子発現の維持と考えられた。
