Research Abstract

本研究領域の研究成果の概要
本領域研究では、ゲノムの安定性を維持するための種々の機構を解明すると共に、それらの欠損によって起こる染色体の不安定化や個体レベルで起こる病態の解析を進めた。
主な成果として:
1.組換え反応の後期過程における大腸菌RuvABCによるHolliday構造の解離の機構を原子レベルで解析し、DNAモーターの動きを可視化した。また真核生物で、組換えの後期過程に関与する新規の遺伝子を4個発見しそれらの機能を解析した。
2.リボゾームRNA遺伝子の重複を安定に維持する種々の仕組みを解明した。
3.真核生物の組換えの前期過程の新しい経路を発見し、構成遺伝子を同定した.
4.酵母をモデルとして、自然に起こるゲノム再編成の制御の機構を解明した。
5.制限酵素と修飾酵素のペアが利己的遺伝子として、ゲノム間を移動し、ゲノム再編成の引金になることを証明した。
6.超好熱古細菌で、多数のゲノム維持に関与する酵素と遺伝子を同定し、人の遺伝病との関連において、重要な貢献をした。
研究成果の取りまとめ
1.平成13年度〜平成17年度 科学研究費補助金 特定領域研究『ゲノムホメオスタシスの分子機構』の研究成果報告書(154ページ)としてわかりやすい形で班員の研究成果の図を入れて解説した。加えて班員の主要な研究成果のうち一流国際学術誌に発表した論文の別刷りを精選論文集(587ページ)としてまとめた。
2.研究経過等の報告書を作成し、事後評価のためのヒアリングで、研究成果を発表し、質疑応答を行った。
3.研究成果公開のため2006年皮紐換え・ゲノム再編ワークショップを共催し、班員が全員研究成果を発表した。
