Research Abstract

ナメクジの嗅覚-味覚連合学習の条件付けにおいて、条件付けの30分前に体腔内に、タンパク合成阻害剤であるanisomycin(0.2mg/g body weight)あるいはoycloheximide(1mg/g body weight)を注射すると、anisomycinの場合、条件付け後二日目以降、cycloheximideの場合、条件付け後一週間目以降の記憶保持に障害が見られた。また脳の[35S]-メチオニンの取り込みを測定したところ、anisomycinは注射後10時間までの間に最大で約90%の取り込み阻害を示したのに対し、cycloheximideでは数時間の間に最大50%程度の阻害しか示さなかった。また、anisomycin投与によって脳でのタンパク合成が十分に抑えられている時間中における、ナメクジの記憶再生そのものは影響を受けなかった。これらの結果から、タンパク合成がナメクジの嗅覚-味覚連合記憶の再生には不要であるが、その長期的な維持には必要であることが明らかになった。さらに条件付け前後のタンパク合成が阻害された場合、記憶の障害が顕著になるまでの時間と、タンパク合成の程度との間に相関が見られることが明らかになった。
嗅覚-味覚連合学習成立後、前脳において特異的に発現誘導を示すことが知られている遺伝子LAPS18が記憶の長期的な維持に関与している可能性を、現在dsRNAによる阻害実験を用いて検討している。
