Research Abstract

癌抑制遺伝子p53の類似遺伝子としてp51、p73を単離した。これらは、癌抑制遺伝子としての機能を有するとともに、細胞、組織の分化発生制御に深く関与していることが判明してきた。本研究を通して、p53ファミリー遺伝子の機能解明、癌治療への応用を目指す。IKKα-ΔNp51Bシグナル経路が皮膚分化に必須である知見が端緒となり、DNA損傷性ストレスに対する細胞応答は、IKKシグナルソームがp51/p73シグナル系路およびNFκBシグナル経路を統合制御することで決定されていることをつきとめた。一過性、弱DNA損傷性ストレスでは、p53のみが安定化し、p21、RR2等の転写を活性化、細胞周期停止、DNA修復を惹起する。一方、持続性、強DNA損傷性ストレスは、p51/p73の安定化をももたらす。このとき、すでに活性化されているp53はp51/p73と協調して、はじめてアポトーシス誘導性プロモーター(BAX、NOXA等)を活性化し、アポトーシスを惹起する。さらに、IKKシグナルソームーp51シグナリングが細胞分化に決定的な役割を担っていることを見出しており、幹細胞や前駆細胞が前癌細胞へ変化することを防ぐチェック機構として機能していると推測している。本年度は、siRNAによる阻害実験にて本現象の検証、扁平上皮形成におけるシグナル経路の機能解明に成功した。さらに、IKKは炎症、自然免疫とも密接に関係しているため、動物個体における解析が必要不可欠であり、現在、遺伝子改変マウスを作成中である。
