Research Abstract

近年のがんの発生発達にかかわるシグナル伝達から、イノシトールリン脂質の関与するシグナル伝達系の細胞のがん化への関与が注目されている。これまで、印環細胞がんの形成メカニズムにPI3キナーゼが関与していることをつきとめた。その活性メカニズムを調べたところErbB3がリン酸化されてPI3キナーゼと結合し活性化していることが明らかとなった。また、下流シグナルとしてはp38MAPキナーゼが関与していることがわかっている。しかし、なぜErbB3が活性化されているのか不明である。そこで、本年度はErbB3活性化のメカニズムについて検討した。印環細胞がんの多くにおいてはErbB2/3が活性化され、ホスファチジルイノシトール3キナーゼ(PI3K)されていて、その結果、細胞間相互作用の喪失、ムチンなどのタンパク質の分泌亢進などの表現型が誘導されていることが明らかになっている。しかしながら、どのようにしてPI3Kが活性化されているかは不明であった。本年度はムチンの一種であり、ErbB2と結合することにより、ErbB2を活性化されるとされていMuc4が実際に印環細胞がん株においてErbB2と結合していること、また、Muc4の分泌を阻害するとErbB2/3の活性化が抑制されることを明らかにした。これらの知見からErbB2-ErbB3-PI3K-Muc4分泌-ErbB2のオートクリンのメカニズムによるErbB2/3の活性化が示唆された。
