Research Abstract

接着分子であるとともにシグナル伝達分子でもあるβ1インテグリンは、癌の浸潤・転移及びに重要な生物学的作用を発揮する。我々がHTLV-I (human T-lymphotropic virus type I)感染T株(Hut78)よりクローニングしたCas-L (Crk-associated substrate lymphocyte type)は、インリンの架橋により強くチロシンリン酸化される細胞内ドッキング蛋白質である。本研究では、テグリンを介するシグナル伝達系の標的分子としてのCas-Lについて解析を行った。
まず、Cas-Lによるシグナル伝達機構とその生物学的意義をさらに解析するため、yeast Two-Hybrid法を用いてHTLV-I感染T細胞株のcDNAライブラリーからCas-Lと結合する可能性のある種々のクローンを単離した。その結果、Cas-Lは、細胞の癌化、癌転移に関与するTGF-βシグナルの阻害蛋白であるSmad6,7に結合すること、Smad6,7のTGF-βシグナル阻害作用を抑制することを明らかにした。また、Cas-LはNF-κB経路を活性化するHTLV-I p40taxに結合し、taxによるNF-κB活性化を抑制することが判明した。さらに、Cas-LとTGF-β・NF-κB両経路との相互作用を検討し、Cas-LがNF-κB経路に必須であるIKK複合体のα,γサブユニットと内因性に結合すること、また、TGF-βreceptorのユビキチン化を行うSmurf-1,2とも結合することを見出した。これらのことからCas-Lは癌細胞増殖・生存・転移等に重要な生物学的意義をもつNF-κB系TGF-β系の双方ともと相互作用する可能性、引いては癌細胞の制御に向けてのひとつの標的分子である可能性が示唆される。
