Research Abstract

網羅的遺伝子発現プロファイルパターンと肝細胞癌の臨床的あるいは病理学的診断との相関を解析することにより、肝細胞癌の診断アルゴリズムの開発を目指し、さらに肝癌特異的な発現を示す遺伝子の機能解析を行い、肝発がんにおける役割を検討した。最大5万個の転写産物について肝細胞癌組織の非癌部と癌部組織においての遺伝子発現プロファイリング解析をおこない、新規の診断、治療の標的分子を探索した。肝細胞癌に高発現するヘパラン硫酸プロテオグリカンGPC3はGPIアンカーされる分子であり、全長分子以外にN末端側が切断を受けた断片sGPC3が血清中に存在することが認められ、ELISAによる血清濃度測定系を樹立した(筆宝、2004)。肝細胞癌切除後の血清sGPC3値が高値の患者群は有意に再発率が高い傾向があり、術後モニタリングのためのバイオマーカーとしての有意義であると考えられた。
発現プロファイル解析に統合するために肝細胞癌に生じている染色体欠失や増幅などのゲノム異常を解明すべく、先に開発した遺伝子発現レベルを染色体領域ごとに評価、表示するアルゴリズムExpression Imbalance Map (EIM)を用いて、遺伝子発現量が増加しているゲノム領域においてのゲノムDNAレベルの変化を定量的PCRによっても確認した。すなわち、発現プロファイルには染色体変異に伴う領域的な偏りを伴うこと報告した(緑川2004)。さらに新規開発したGenome Imbalance Map (GIM)ソフトウェアを用いてアレル別のコピー数解析が進行中である。
