Research Abstract

リボザイムは、配列特異的にRNAを切断することが可能な酵素であり、癌やウイルス疾患の原因遺伝子を標的とした遺伝子治療剤としての可能性が期待されている。本研究では、基質認識部位をランダマイズしたリボザイムライブラリーを用いた新規機能遺伝子探索法(ジーンディスカバリーシステム)を開発し、それを用いて新たな癌標的分子を同定することを目的としている。今年度は、リボザイムライブラリーを用いたジーンディスカバリーシステムを用いて癌転移・浸潤に関わる機能遺伝子の探索を培養細胞レベルとマウスの個体レベルで試みた。癌の浸潤のモデル系であるゲルインベージョンアッセイ法を用いて機能遺伝子探索も行った結果、GemGTPaseやPEBP2などの浸潤能を抑制可能な機能遺伝子を同定することに成功した。また転移能を持たないメラノーマ細胞にリボザイムリブラリーを導入した後にマウスの尾部に注入して、肺への黒色腫の形成能をもとに機能遺伝子の探索をおこなったところ、リボザイムリブラリーに起因する多くの黒色腫を持つマウスが多数得られ、STIM1やpolg2などの標的遺伝子を同定することに成功した。これらの結果は、リボザイムライブラリーを用いた機能遺伝子探索法が個体レベルでも機能していることを示している。またこれらの同定された遺伝子は、癌の転移を抑制する機能を有する可能性が考えられ新しい癌分子標的となりうることも考えられる。また乳癌細胞で過剰発現しているCyclin D1とHst1を標的としたマキシザイムを構築して各種組織の癌細胞や正常細胞への影響を検討した結果、マキシザイムは乳癌細胞において高い特異性と癌遺伝子の発現抑制を示すことがわかった。このことから正常細胞には副作用を及ぼさない特異性の高いマキシザイムは、将来的な各組織の癌分子標的治療への応用に期待できると思われる。
