Research Abstract

白血病に対する新たな免疫遺伝子療法の開発を目的として研究を遂行し、以下の成果を得た。1)骨髄腫細胞はリンパ腫細胞と比べて、WT1特異的細胞傷害性T細胞(CTL)によるパーフォリン依存性細胞傷害感受性が高いことが明らかとなった。2)CD4陽性およびCD8陽性CTLにおけるパーフォリン発現と細胞周期との関連を調べたところ、CD8陽性CTLでは細胞周期に関わらず恒常的に発現が認められたが、CD4陽性CTLではG2/M期に転写が促進することが明らかとなった。この違いは、STAT5のパーフォリンプロモーターへの結合の差によることが判明した。3)腫瘍細胞に強い細胞傷害性を示すWT1特異的CD8陽性CTLクローンからT細胞レセプターα鎖およびβ鎖遺伝子をクローニングし、正常CD4陽性およびCD8陽性T細胞に遺伝子導入することによって、白血病細胞に対してHLAクラスI拘束性に細胞傷害性が獲得されることが明らかとなった。ただし、CD4陽性T細胞による機能発現には標的細胞上のHLAクラスIIとCD4分子との結合が必要であることも明らかとなった。4)HLA-DP5拘束性CD4陽性T細胞が認識する新たなWT1エピトープを同定した。このペプチド特異的CD4陽性T細胞はWT1特異的に白血病細胞を殺傷した。5)これまでの基礎研究成果をもとに、WT1とhTERT由来ペプチドを用いたがんペプチドワクチンの第I相臨床試験を遂行している。グレード2以上の有害事象は認められず、一部の症例に臨床効果が得られている。6)新たな免疫不全マウスにヒト造血幹細胞を移植することによってヒト免疫系を構築したマウスを作製した。今後、ヒトにおけるワクチン開発に有用であると思われる。
