Research Abstract

急性骨髄性白血病に観察されるinv(12)(p13q13)より3'-RACE法を用いて新規キメラ遺伝子TEL/PTPRRをクローニングした。得られたキメラcDNAはTEL遺伝子の第4エクソンにPTPRR遺伝子の第7エクソンが融合していた。PTPRRはTEL転座の相手遺伝子としては初めての受容体型チロシン・ホスファターゼ遺伝子であった。TEL/PTPRR遺伝子から転写されるmRNAには10種類のisoformが存在し、短縮型TELとTEL/PTPRRキメラに翻訳されると考えられた。野生型TELは核内に存在するが、短縮型TELおよびTEL/PTPRRキメラは細胞質内に存在していた。これらの異常分子は野生型TELとヘテロダイマーを形成することにより、その核移行を抑制すると推測された。短縮型TELとTEL/PTPRRキメラには野生型TELに認められるコリプレッサーmSin3Aとの結合能が保存されていた。また、短縮型TELとTEL/PTPRRキメラはETS結合部位に対するDNA結合能および転写抑制能を示さなかったが、野生型TELの転写抑制能をドミナントに阻害した。TEL/PTPRRキメラにはボスファターゼ活性は認められなかった。TEL/PTPRRはGM-CSF依存性に増殖するヒト白血病細胞株UT7/GMをGM-CSF非存在下でも増殖させた。短縮型TELには同様の機能は存在しなかった。さらに、短縮型TELあるいはTEL/PTPRRキメラを発現させたUT7/GM細胞はGM-CSF除去後もSIAT3のリン酸化状態を維持していた。inv(12)型白血病ではがん抑制因子TELが機能的に失活し、STAT3シグナルが恒常的に活性化していると考えられた。
