Research Abstract

肝癌においてはレチノイド核内受容体(RXRα)がErによるリン酸化を受け、その機能を失うため、癌細胞がレチノイド不応性に陥っていることを明らかにした。またリン酸化RXRαは、ユビキチン/プロテアソーム系による分解を回避して分解が遅延し、癌細胞内に蓄積してdominant negativeに作用するため、正常なレチノイドの細胞増殖抑制作用を阻害し、細胞増殖を亢めて、肝発癌に関与することを明らかにした。一方、我々の開発した非環式レチノイドは、Erk系を阻害し、RXRαのリン酸化を抑制してその機能を回復させる働きを有することを明らかにした。RXRαは様々な転写活性に関与するmaster regulatorであることが知られている。そこでRXRαリン酸化の他の癌腫における発癌過程への関与をも検討し、大腸癌においてはPPARγと、急性前骨髄性白血病においてはRARとヘテロダイマー形成能が失われ、肝癌同様にRXRαのリン酸化が発癌過程に密接に関連していることを明らかにした。
