Research Abstract

ヒトTLRに対するモノクローナル抗体はTLR1,TLR2,TLR3,TLR6を作製し終えた。TLR4の抗体は三宅博士(東大医科研)より供与された。樹状細胞上のTLR分子複合体の組成を明らかにする企画で以下の知見を得た。1、微生物成分は結合とシグナルグを別々のレセプターで分担する。現在、各TLRの相互関係とリガンド応答性の機能解析中である。
BCG-細胞骨格成分(CWS)ではintelectinなどのC-type lectinが結合レセプターとして機能する。BCG-peptidoglycan(PGN)ではpeptidoglycan-recognition protein-I(PGRP-I)が結合レセプターとして機能する。シグナルはTLR2とTLR4によって伝えられる。この点は抗体阻害実験によっても確認できた。また各TLRの発現細胞株を用いたレポータージーンアッセイでも確認した。従って、ヒト、マウスに共通の微生物peptidoglycan認識系がtwo receptorsとして存在することが証明できた。
TLR3につい新規リンガンドをdsRNAと同定した。外国グループとの競合で出版が遅れているが新規のシグナル系、ウイルス感染との関係を解析していく予定である。ノックアウトマウスの系と比較し、ヒトTLR/樹状細胞に特有な微生物応答反応を尊重していく。
ヒト樹状細胞の応答には個体差が見出される。患者検体を用いた解析では、当センターの個人情報・倫理委員会において連結可能匿名化が認められた。
