Research Abstract

In order to understand aquatic environments in Southeast Asia biologically, we performed development of DNA microarray for analyzing bacterial community structure and identification of environmental stress response genes of fish and alga.
1. Bacterial community structures in rivers and canals in Bangkok were analyzed, and dominant bacterial groups were identified.
2. Changes and similarities of bacterial community structures in a microcosm were monitored by DNA microarry targeting 16S ribosomal RNA genes.
3. Expression of the metallothionein gene in the fish culture cells was induced by treatment with cadmium, zinc, arsenic, or t-butyl hydroperoxide. The expression level was depend on their concentration and treatment time.
4. Expression of the heat shock protein gene (hsp70) in the fish culture cells was induced by arsenic treatment.
5. Phytochelatin synthesis was induced strongly, by activation of glutathione synthesis pathway with heavy metal treatment in marine green alga, Dunal3ella tertlofecta.
6. Phytochelatin can reduce the oxidation stress caused by various environmental stresses in Dunaliella tertiolecta.
