Research Abstract

Heart is positioned in center of life support. In outbreak, the heart serves as specialization quickly.It takes a role, and to push forward the specialization formation of other organs. However, to a heart,there is not regenerative ability. As for the treatment of function compensation of disorcter cardiac muscle, there is only heart transplantation.
We try establishment of HIF- 1 (HIFrhipoxia-inducible factor) gene therapyin order to revitalize this disorder cardiac muscle. We let them promote angiogenesis by gene induction. And we let them regenerate the cardiac muscle cells which seems to be lifeless in ischemia.
We tried manufacture of the ischemic heart muscle model with mini-pig. We mounted center of left coronary artery circumflex with Ameroid constrictor. In miniature pig of weight from 20 kg to 30 kg,Ameroid-size-1.9 were optimum size. In this ischemic heart model, we performed an experiment of derivation of angiogenesis by HIF- 1 gene therapy.
We proved that angiogenesis promoted it by this gene therapy in selective coronary arteriography.We used colored microsphere and confirmed increase of regional myocardial blood flow. Thin bridge collateral near Ameroid increased.
It was confirmed that HIF- 1 gene therapy to ischemic heart promoted angiogenesis.
