Research Abstract |
To develop an anti-obesity vaccine for domestic animals and a safety low fat meat production, we investigated 1)the regulatory mechanism of adipocyte differentiation on animal preadipocyte, 2)screening of functional nutrient for the inhibition of adipocyte hyperplasa, and 3)the effect of intraperitoneal administration with inhibitor of adipocyte proliferation on adipose tissue weight. 1)The regulatory mechanism of chicken adipocyte differentiation To induce the chicken pre-adipocyte differentiation, we firstly clarified that the addition of an exogenous fatty acid mixed with hormones (Ins, DEX, IMX), which induce the differentiation of mammalian adipoocyte, was essential for chicken adipogensis. In addition, it is determine that PPARγ is master regulator of chicken adipocyte differentiation during adipogensis. 2) Screening of functional nutrient for the inhibition of adipocyte hyperplasa The adipocyte proliferation declined with the addition of the mixture of insulin. 3-isobutyl-1-methylxanthin, dexamethason and oleic acid, and completely inhibition of cell growth occurred at the concentration of 20μg/ml insulin, 0.5mM 3-isobutyl-1-methylxanthin, 500nM dexamethason and 300μM oleic acid in the media. 3)The effect of intraperitoneal administration with inhibitor of adipocyte proliferation on adipose tissue weight In chickens with single intraperitoneal injection of these hormone and fatty acid mixture made emulsion with triolein, adipose fat pad weight was significantly lower than the control chickens whereas the body weight decreased to a small extent. In addition, the administration of our anti-obesity vaccine regulated in adipocyte hyperplsia. Unfortunately, it was not still known the powerful nutrient, which was easily accepted by consumer, for the regulation of adipocyte hyperplasia, but the new manipulation technique reducing fat deposition, called anti-obesity vaccine, was developed by the control of adipogensis in chickens.
