Research Abstract

It was suggested that the charge transfer rate through the base molecules in a double-helix DNA molecule is strongly dependent on the base-pairs alignment in a biochemical analysis. In this study we directly observed the energy spectra of the electron-density of states just on the each base-site with using scanning tunneling microscopy/spectroscopy. We compared the spatially-resolved tunneling spectra of DNA(poly(dA-dT) ・ poly(dA-dT)) with those of DNA(poly(dG-dC) ・ poly(dG-dC)) and found out that the latter has the higher electron energy for the shallowest DOS peak in the valence band. This fact means that a (G-C) pair performs as a lower barrier for holes compared with an (A-T) pair. This conclusion is consistent with the elucidation by the biochemical approach.
