Research Abstract

PGPF isolates from zoysiagrass rhizosphere that promote plant growth were tested for their ability to induce, systemic resistance in cucumber plants against Colletotrichum orbiculare, the anthracnose pathogen. Second true leaves of cucumber plants were treated with culture filtrates (CFs) of these fungal isolates and 24 hrs later third true leaves were inoculated with pathogen. Induced plants significantly reduced the total lesion area compared to uninduced control plants. Value of lignin deposition at points of attempted penetration by the pathogen in induced plants were significantly higher than in control plants. Lignin was formed 24 hrs after pathogen treatment. The elicitor activity of CFs was evaluated in cucumber fruit disks using a luminol-dependent chemiluminescence assay. The CFs of the PGPF isolates elicited a conspicuous superoxide generation from cucumber fruit disks when compared with control. Experiments were also done to determine whether these isolates could activate peroxidase and chitinase genes in cucumber plants or not. Expression of peroxidase and chitinase genes were observed 2 and 4 hrs after each CF-treatment, respectively, suggesting occurrence of priming effect in the cucumber plants. When the hypocotyls of each CF-treated cucumber was challenge inoculated with C. orbiculare, the activation of these genes was more rapid and prominent.
