Research Abstract

The hypothalamic dorsomedial nucleus (DMH) is suggested to be a final relay site for the afferent of milk-ejection reflex (MER) [1]. Existence of prolactin releasing peptide (PrRP) - immunoreactive cells [2] and cells expressing PrRP-mRNA[3] in the DMH and synaptic contact of PrRP-IR terminals with oxytocin (OT) cells was reported [2]. It was also shown that intracerebroventricular injection of PrRP stimulated oxytocin release in rats[4]. Experiments were done to test the effect of PrRP on the electrical activity of OT cells in the supraoptic nucleus (SON). In rat hypothalamic slice preparations containing supraoptic nucleus, OT cells were unresponsive to peptides. In lactating rats, whereas lateral ventricular (LV) injection of PrRP (20-30 nmol) were ineffective, a hundred nmol of the peptide increased basal activity and amplitude of bursting activities of oxytocin cells. Cells responded to LV injection of PrRP were unresponsive to direct application of the peptide by pressure ejection from the recording electrode. Results suggest that PrRP may modulate activity of OT cells through its action on the area other than SON.
