Research Abstract |
Recently we identified the 182 amino acid NS2 protein as a protein of 〜22kDa in influenza C virus-infected cells. In this study, it was demonstrated that the NS2 protein of influenza C virus was present in purified virions as follows. 1. Western blot analysis with anti-NS2 serum detected the NS2 protein in egg-grown influenza C/Yamagata/1/88 virions purified by centrifugation through a linear gradient of 5%-40% potassium tartrate. NS2 was also present either in purified virions of other strains grown in eggs or in purified C/Yamagata/1/88 virions grown in other host cells(MDCK or HMV-II). 2. The NS2 protein was detected in influenza C virions purified by using the binding activity either to the rat red blood cells or to anti-Hemagglutinin-esterase(HE) monoclonal antibody-conjugated protein A sepharose. 3. The NS2 protein associated with influenza C virions purified by centrifugation through a linear gradient of 5%-40% potassium tartrate was completely resistant to the treatment with 200 μg/ml of TPCK-trypsin for 30 min at 37℃. 4. These data indicate that the NS2 protein of influenza C virus is present inside the viral envelope.
