Research Abstract |
The role of Cbl in FcεRI-induced signaling cascade in mast cells were examined, especially focused on the role of proline rich region in Cbl. Proline (543-548) were mutated by a cite direct mutagenesis, and the mutated Cbl was expressed in mast cells. The levels of tyrosine phosphorylation of Syk and association between Syk and γ chain of FCεRI were unchanged. However, activation of transcription factor NF-κB and histamine release were slightly enhanced in cells transfected with the mutated Cbl compared with that in cells transfected with wild type Cbl. These results collectively indicated that proline rich region of Cbl regulates the FCεRI-induced signaling cascade in mast cells.
