Research Abstract

We investigated the relationship between proliferation index, expression of cell cycle inhibitors and telomerase activity in brain tumors. In meningiomas, telomerase activity was found only in high grade tumors. We found correlation between the expression of hTERT and proliferating index (MIB-1). However, immunohistochemical study revealed that even in benign tumor hTERT was expressed in several cells that were MIB-1 positive. We demonstrated that these hTERT positive cells were definitely meningioma cells by double staining with EMA and hTERT. Tumors with high rate of hTERT positive cells are usually atypical or anaplastic meningiomas and exhibited a high recurrence rate. Similar findings were also found in gliomas.
Expression of p27 was frequently observed in meningiomas. The relationship between the proliferation indices and frequency of p27 expression was not distinct. However, we found correlation between cellularity and p27 expression . p27 was also expressed in perinecrotic area. Expression of p21 was relatively rare and correlated with the proliferation index. On the contrary,expression of p16 adversely correlated with proliferation index. Calcified tumors often showed p16 positive cells, and they usually lack hTERT expression. However, we could not demonstrated the correlation between p16 expression and telomere length.
