Research Abstract |
The aim of this study is to prove our hypothesis that retinoid signaling plays a central role in regulation of endochondral ossification by control of expression and function of Indian hedgehog (lhh) and Cbfa1 transcriptionactor. We have found that (1) retinoid signal is activated in the penchondrium close to hypertrophic zone, but not * the other parts of cartilage including the penchondrium, (2) Implantation of beads soaked with retinoids in the deleveloping cartilage induces cbfa1 expression, and implantation of beads soaked with antagonist of retinoids inhibits hypertrophy of chondrocytes, (3) stimulation of hypertrophy of chondrocytes induced by retinoid is completely inhibited by overexpression of dominant negative form of Cbfa1 in vitro, but retinoid antagonist does not disturb the stimulatory action of Cbfa1 on hypertrophy of chondrocytes and (4) retinoid up-regulates expression of lhh in immature chondrocytes while it down-regulates expression of lhh in mature cells. It is suggested that retinoid signal stimulates chondrocyte maturation through up-regulation of cbfa1 expression and that retinoid signal down-regulates lhh expression, and thereby diminish lhh-negative control in chondrocyte maturation. Retinoid signalnight be a key in regulation of the balance between suppression and induction of chondrocyte hypertrophy during endochondral ossification.
