Research Abstract |
Objects: We designed to elevate therapeutic efficacy of radiation by addition of glycerol on an oral squamous cell carcinoma cell line (Ca9-22) bearing mutation in a tumor suppressor gene, p53 (mp53). Methods: To examine the effectiveness of glycerol in radiation-induced lethality for Ca9-22 cells, we performed colony formation assay. Apoptosis was analyzed with Hoechst 33342 staining, Western blotting and DNA ladder formation assay. Ca9-22 cells became radiosensitive when glycerol was added to culture medium before X-ray irradiation. Results: In accordance with the cell survived rate, apoptosis (apoptotic body and DNA fragmentation) was induced by X-rays irradiation after glycerol treatment while there was little apoptosis induced by X-ray irradiation or glycerol addition alone. Bax protein was accumulated after combined treatment of X-ray irradiation plus glycerol, but not after X-ray irradiation. Conclusions: These findings suggested that pre-treatment with glycerol might be effective in therapy against mp53 cancer cells.
