Research Abstract |
This study was undertaken to investigate the gene expressions of IGF-I, FGFs and MyoD following denervation and compensatory overload. 10 wk-olds Wistar strain rats were used in this study. The rats were divided into 4 groups, innervated and compensatory overloaded group (NM-OL), innervated and sham operated group (NM-Cont), denervated and compensatory overloaded group (DN-OL), and denervated and sham operated group (DN-Cont). IGF-I, PGFs and MyoD mRNA expressions as well as weights of plantaris muscle were measured 3, 7 and 21 days, and myosin heavy chain isoforms were measured 21 days after the treatments. At 21 days, type I myosin heavy chain isoform in NM-OL and DN-OL were significantly increased as compared with NM-Cont and DN-Cont, respectively. At 3, 7 and 21 days, muscle weights of the plantaris in NM-OL and DN-OL were significantly increased as compared with NM-Cont and DN-Cont, respectively. At 3, 7 and 21 days, FGF-2 mRNA and FGF-6 mRNA expressions in NM-OL and DN-OL were increased as compared with NM-Cont and DN-Cont, respectively. At 3 and 7 days, IGF-I mRNA and MyoD mRNA expressions in NM-OL and DN-OL were increased as compared with NM-Cont and DN-Cont, respectively. The plantaris muscles were strophied by the denervation. The expressions of FGF-2 mRNA and FGF-6 mRNA in DN-Cont and DN-OL were lower than the NM-Cont and NM-OL, respectively, however, the expressions of IGF mRNA and MyoD mRNA in DN-Cont and DN-OL were higher or unchanged. Thus it would seem to be likely that IGP-I, FGFs and MyoD were completely associated with the morphological changes of plantaris muscle following the denervation and compensatory overload.
