Research Abstract

ニホンナシでは交配により7つのニホンナシの雌しべ側S遺伝子産物としてS-RNaseが同定されている.S-RNaseにはS遺伝子間でアミノ酸配列を異にする1つの超可変領域(HV領域)が存在し,花粉と雌しべの自他認識に関与しているとされる.HV領域とHV領域に挿入されたイントロンの塩基配列をもとにS_1-S_7遺伝子を識別できるPCR-RFLPシステムを用い,主要品種遺伝子型の推定を進めた.前年度までに'新興'と'新星'にS_1-S_8-RNaseとは異なるS_9-RNase cDNA断片をクローニングし,それらの遺伝子型がS_4S_9であること示している.
本年度はS_9-RNase cDNAの全塩基配列を決定するため、'新興'と'新星'の花柱からmRNAを抽出し,5'-RACEによりS_9-RNase cDNAの5'末端の塩基配列を決定した.S_1-S_9-RNaseの塩基配列に基づき新たにリバースプライマーをデザインし,ゲノムDNAからS_9-RNase断片を増幅し,塩基配列を決定した.S_1-S_9-RNaseのS_9-RNaseのHV領域とイントロンを配列も基づき,S_9-RNaseを特異的に切断する制限酵素BstBIとS_2-RNaseを特異的に切断する制限酵素AflIIを選択した.
'FTQQYQ'と'anti-(I/T) IWPNV'プライマーを用いたPCRによりゲノムDNAから増幅したS_1-S_9-RNase断片を9種類のS遺伝子特異的な制限酵素(Sfc I, Afl II, Ppu MI, Nde I, Alw NI, Hinc II, Acc II, Nru I, Bst BI)で切断することにより,S1-S9の対立遺伝子を有するニホンナシ品種のS遺伝子型を同定できるPCR-RFLP(S1-S9)システムを開発することができた.
