Research Abstract

1.生体内で抗酸化作用を発揮する天然化合物の検索
解毒酵素誘導能のスクリーニングから、東南アジアにおいて著名なハーブ、レモングラスの主要な精油成分であるシトラール及び合成抗酸化剤であるブチルハイドロキノンに、強力なグルタチオン転移酵素誘導活性を見出し、新しい内因性細胞防御因子として評価できた。その活性発現には、細胞内の蛋白質性SH基とのMichael反応が必須であり、転写因子Nrf2が関与することが示唆された。これまでに、ブチルハイドロキノンの解毒酵素誘導に活性酸素生成反応が重要であることが他のグループによって提唱されていたが、活性酸素は生成するが求電子性は欠如した誘導体であるジブチルハイドロキノンを用いた詳細な実験から、活性酸素の関与を否定する結果も得られた。一方、シトラールについては、マウス皮膚での炎症性酸化ストレスを有意に抑制し、新しい抗酸化性発がん予防剤の候補になり得るものと評価できた。その他、熱帯産ショウガ由来ゼルンボン、合成フェルラ酸誘導体、黒酢由来ジヒドロフェルラ酸などに顕著な抗炎症作用とともに、活性酸素及び活性窒素種の産生阻害作用を見出した。以上の様に、様々な素材に生体内で抗酸化作用を発揮する化合物を求め、単離精製、活性評価、誘導機構の解析を行い、数種の新規活性物質を見い出した他、これまでにはない生体内での活性酸素レベル制御ストラテジーを提唱するに至っている。
