Research Abstract

末梢神経移植による視神経再生の際に、神経細胞とシュワン細胞の間において、本来は同種細胞の間にのみ見られるはずのtight junctionとgap junctionが形成される.この特異なjunctionの神経再生における役割を様々な手法を用いて明らかにし、接着構造蛋白の遺伝子発現をin vivoの視神経再生系において操作し、視神経再生の動態を解析した.
培養シュワン細胞を用いた人工移植片の作成であるが、生後1-3日の新生仔ラット脊髄後根神経節よりシュワン細胞を離離・培養し細胞外基質、神経栄養因子と共に物質過性チューブに充填して人工移植片を作成する条件を設定した.またかかる移植片を成体ラットの切断坐骨神経及び視神経に移植して再生を確認した。occludin並びにconnexin32,43の導入遺伝子を細胞レベルで培養シュワン細胞に導入し移植を行った.また網膜神経節細胞への遺伝子導入方法はelectroporation法を改変し実際の生体眼における遺伝子導入の実現にむけて独自の開発を行なった.全域で平均50%、網膜中心部で70-80%という高率の遺伝子導入に成功し、組織傷害や副作用が起きないことを確認し、同様に遺伝子を網膜神経節細胞に導入した.視神経再生率・再生速度は硝子体に注入したコレラトキシンサブユニットBの順行性標識した再生視神経の追跡によって評価した。シュワン細胞に上記の遺伝子を導入した場合、また網膜神経節に導入した場合、共に遺伝子を導入しなかったものとの比較においてわずかながらの視神経再生率・再生速度の向上が見られたものの統計的な有意差は認められなかった。
