Research Abstract

表面プラズモン共鳴現象を応用した,生体内分子間相互作用解析装置を用いて,定量的テロメラーゼ活性の評価法telomeric repeatelongation (TRE) assay を開発した.TRE assayはBio sensor chipと呼ばれる金薄膜のコートされた1cm^2程のチップ上に,G-quartet構造を持った50bp程のDNA断片を固定化する.次に,テロメラーゼ抽出物ををロードし,結合・伸長・解離・再生を行う.得られたセンサーグラムから伸長の結果生じた重量変化をe-valueとした.本法の特徴は (1)PCR法を用いていないので,従来のTRAP法で生じていたPCR inhibitorの混入によるfalse-negativeがない
(2)塩基の伸長速度を直接計測できる
(3)電気泳動を必要としないone-step操作なので,ピペット操作や泳動操作の際に生じる誤差がない.
(4)デンシトメータあるいはELISAによる計測の必要がない.
(5)短時間にnon-labelingで解析可能である.
などの利点があった.
TRE assayはnon-labelingで,これまでのTRAP法による阻害効果の検討時間を1/50程に短縮する事が可能であり,本法を用いたテロメラーゼ阻害物質のスクリーニングが可能である.TRE assayは新規テロメラーゼ阻害剤の発見に効果を発揮し,テロメラーゼを標的とした分子標的治療法の開発に大いに貢献すると思われた.
