Research Abstract

我々はyeast two-hybrid screeningによって,NEDD8に結合する70kDaの新規タンパク質を単離することに成功し,NUB1(NEDD8 Ultimate Buster-1)と命名した.NUB1を細胞内で過剰発現させるとNEDD8の減少が観察されたのに対し,Ubや同じUb-like proteinであるSentrin-1(SUMO-1),Sentrin-2に対しては全く影響を及ぼさなかった.我々はNUB1の特異抗体を作製し,ほとんどの臓器で70kDaの単一バンドとして発現していること,細胞内では核に優位に限局していることを明らかにした.さらに,培養細胞にinterforonを作用させるとNUB1発現が亢進することを明らかにした.NUB1によるNEDD8分解活性がproteasomeまたはprotease inhibitorにより抑制されるか否かについて検討したところ,calpain inhibitorであるLLMでは抑制されなかったのに対し,proteasome inhibitorの作用をもつLLnL,MG-132,さらにproteasome特異的inhibitorであるclasta-lactacystin β-lactoneでNUB1の活性が抑制された.NUB1は26Sproteasome分画には存在するものの,20S分画には存在しないことから,調節ユニットであるPA700と結合していることが示唆され,実際にPA700の構成分子のひとつであるS5aと結合することを明らかにした.また,我々はubiquitinと交差反応を示さないNEDD8特異抗体を作製し,免疫組織化学的に臓器での発現を詳細に報告し,NEDD8がubiquitin陽性を示す病的封入体においても蓄積していることを明らかにした.
