Research Abstract

世界人口の半数以上のヒトが感染しているヘリコバクター・ピロリは持続感染の結果、一部のヒトに、慢性胃炎、胃・十二指腸潰瘍、胃癌、MALTリンパ腫などの発症を引き起こす。ヘリコバクター・ピロリのうち、cag Pathogenicity Island (PAI)を持つヘリコバクター・ピロリが宿主の胃上皮細胞のNF-κBを顕著に活性化し、ピロリ菌による炎症との関わりが指摘されている。従って、ピロリ菌によるNF-κBの活性化機構を解明することは、臨床的に重要であると思われる。
本研究では、ピロリ菌により引き起こされるNF-κBの活性化がいかなる動物細胞においても認められるのかどうか調べてみた。胃上皮由来株化細胞MKN45細胞、AGS細胞、AZ-521細胞、および腎臓上皮由来株化細胞293T細胞、Cos-7細胞を、ピロリ菌にて刺激をおこない、NF-κBの活性化が誘導されるかどうかレポーター遺伝子を用いて解析をおこなった。その結果、ピロリ菌によるNF-κBの活性化がMKN45細胞、AGS細胞、293T細胞では認められ、AZ-521細胞、Cos-7細胞では認められなかった。以上の結果は、ピロリ菌によるNF-κBの活性化は細胞特異性があること、また、すべての胃上皮細胞においてNF-κBが活性化されるのではないことがわかった。
