Research Abstract

NO-4007(LPS誘導体)、INF-γ、meglumine antimoniateのcombination投与による抗リーシュマニア作用について
(i)リーシュマニア感染BALB/cマウスに対して;2×10^6のLeishmania amazonensis promastigotesを背部皮膚に接種2週間後より薬剤投与し、10週間病変発現を観察したところ、meglumine antimoniate+IFN-γ連日投与群で病変の発現を完全に抑制した。meglumin antimoniate連日+ONO-4007週3回投与群はmeglumine antimoniate連日+IFN-γ週3回投与群と同程度に病変発現を著明に抑制した。また薬剤単独投与よりcombination投与の有効性が示唆された。
(ii)リーシュマニア感染J774cellに対して;Leishmania amazonensisまたはmajorを感染させたJ774cellに薬剤を添加し48時間後にその上清のTNF-αをELIZA法にて測定した。またJ774cellを電顕的に観察した。その結果、ONO-4007投与群でのみTNF-αの著明な誘導を認めた。J774cell内の原虫数は薬剤投与群では著明に低下しており、parasitophorous vacuoleは薬剤投与群全てで形成抑制がみられた。これらの結果はONO-4007の他剤とは異なる原虫排除機構を示唆しており興味深い。
