Research Abstract

フォンビルブランド因子特異的切断酵素(VWF-CP)の精製をFujikawaらの方法をもとに改変し行った。蛋白精製の出発材料は有効期限切れの新鮮凍結血漿(fresh frozenplasma、FFP)である。これをDFPで処理し、その後lysin-sepharoseカラムを通して、まず血漿中のplasminogenを除去した。以後、硫安沈澱、butyl-sepharoseカラム、Q-sepahroseカラム、superdex200カラムにてVWF-CPを部分精製した。これを抗原として、マウスとウサギに免疫し、前者からはモノクローナル抗体を、後者からはポリクローナル抗体を作成した。モノクローナル抗体は、ELISAおよびウエスタンブロットでスクリーニングを行ったが、VWF-CPに特異的に反応するモノクローナル抗体は得られなかった。しかし、ポリクローナル抗体は、ウエスタンブロットでVWF-CPと強く反応し、また酵素活性を阻害することも確認した。阻害力価は、ポリクローナル抗体IgG 1mgあたり8.9 Bethesda Unitであった。なお、1 Betheda UnitはVWF-CP活性を50%阻害する力価である。今後、このポリクローナル抗体を用いVWF-CPを精製するか、遺伝子発現したVWF-CPを用いてマウスにするかして、再度モノクローナル抗体の作成を試みる予定である。
