Research Abstract

角膜上皮欠損後の上皮細胞遊走過程でのケラタン硫酸プロテオグリカンのコア蛋白質であるルミカンの役割に関する研究と、関連する眼創傷治癒過程での様々な細胞の挙動に対するルミカンの影響を検討した。角膜上皮細胞の創傷治癒過程では、先端の上皮細胞は、上皮-間葉系移行に類似した挙動をとり、ビメンチンを一過性に発現することが以前に確認出来ていたが、さらに上皮-間葉系移行でのルミカンの役割を検討するために他の細胞も用いた。水晶体上皮は、他の細胞の混入なく、創傷治癒過程での上皮-間葉系移行を研究できることから、まず、創傷後の水晶体上皮細胞でのルミカン発現を蛋白質レベルで検討したところ、水晶体上皮細胞も角膜上皮細胞と同様に創傷治癒過程で一過性にルミカンを発現した。さらに、ルミカンノックアウトマウス(Lum)の水晶体外傷での上皮-間葉系移行を検討した結果、Lum-/-マウスで、Lum+/-マウスと比較して、優位に遅延した。このことを受けて、角膜上皮細胞の治癒過程での先端細胞のビメンチン発現を検討したが、角膜上皮細胞の遊走はルミカン欠損で遅延するものの、ビメンチン発現には優位な変化は観察されず、ルミカンの役割の面角膜上皮細胞と水晶体上皮細胞での差異が確認された。一方、角膜穿孔外傷後の治癒過程をLum-/-マウスで、Lum+/-マウスと比較したところ、高額顕微鏡レベルでは差異は検出されなかったが、透過電子顕微鏡レベルでは、創傷に伴う実質コラーゲン線維の乱れの正常化はLum-/-マウスで遅延していた。
