Research Abstract

The effect of dioxin and endocrine disrupters on adrenal hormone in mice were examined in this study.
Most studies of endocrine disrupting chemicals have focused on the sex hormone, and (anti-)estrogenic and (anti-)androgen activities in numbers of chemicals are screening. However, endocrine glands other than a reproduction system might be disrupted by environmental pollutants. Therefore, we examined the effects and toxicities of dioxin and environmental pollutants on adrenal cortex hormone, which significant hormone for life maintenance.
The steroid hormones of adrenal cortex is generated by specific cytochrome P450 isozymes, P450scc, P45011b, and P450c21. The effect of 2, 3, 7, 8-tetrachlorodibenzo-p-dioxin (TCDD) and methylchranthrene (MC) on these enzymes were examined. TCDD and MC were administrated to C57BL/6J Jcl mice by gavage.
Consequently, when Western blot analysis using each antibody were performed, change of the amount of enzyme proteins were not observed. Then, the enzyme activities were more sensitive than Western blot, these P450 activities were measured. When P45017a and 11b activities were measured in adrenal microsomes of mice. The activities in TCDD treated mice were lower than vehicle control. These results supported the previous reports that corticosteron level in TCDD treated rats were lower than control.
In this study, these facts supported that cytochrome P450s which concern the generation of adrenal cortex hormone were disrupted by TCDD.
