Research Abstract

【目的】臍帯血より心筋分化能を有する可能性のある細胞を単離し、増殖し、分化誘導を試みた。【方法】(1)臍帯血よりauto MACSを用いて、CD45陰性、CD29陽性、CD34陰性、CD44陽性の細胞群を単離し、ファイブロネクチン塗布培養皿上に接着させた。(2)この細胞を培養し、細胞の増殖曲線を作成した。(3)この細胞の表面抗原をFACS解析した。(4)この細胞をインスリン存在下で培養し、脂肪細胞への分化を観察した。(5)この細胞を0.1mMデキサメサゾン、0.05mMアスコルビン酸-2リン酸、10mMβ-グリセロフォスフィト存在下で培養し、骨芽細胞への分化を観察した。【結果】(1)臍帯血より上記表面抗原の抗体を利用することにより、付着系の間葉系幹細胞が回収できた。この細胞はin vitroである程度増殖させることが可能であった。(2)増殖曲線は一週間で3倍程度の増加速度であった。(3)インスリン存在下ではこの細胞は脂肪細胞に分化した。(4)デキサメサゾン、アスコルビン酸-2リン酸、β-グリセロフォスフィト存在下ではこの細胞は骨芽細胞に分化した。フォンコッサ染色陽性の細胞が観察された。【結論】臍帯血細胞中には間葉系の幹細胞が存在した。この細胞が心筋細胞に分化するかはさらなる研究が必要と考えられた。
