Research Abstract

I)プロテアソーム阻害剤(MG-132)に対するHsp27とαBクリスタリンの応答を、ヒトグリオーマ細胞U373MGおよびU251MGを用いて解析し、以下の結果を得た。
(1)MG-132で誘発される両シャペロンの誘導および不溶性分画への蓄積とユビキチン化蛋白質の蓄積は、細胞内封入体の形成をもたらし、細胞死を引き起こした。
(2)しかし、シクロヘキシミドなどの蛋白合成阻害剤を共存させると、熱ショック因子(HSF)の活性化が阻止され、不要性分画への上記蛋白質の蓄積と封入体形成が阻止されて細胞死が一時的に防御された。
(3)逆に、MG-132によるHSFの活性化を促進する作用のあるlipoxygenaseの阻害剤、nordihydroguaiaretic acid (NDGA)を共存させると、両シャペロンの不溶性分画への蓄積とユビキチン化蛋白質の蓄積が増加した。NDGA単独では上記の作用は認められなかった。
以上の結果より、プロテアソーム阻害剤によるαBクリスタリンおよびHsp27の不溶性分画への蓄積はHSFの活性化が密接に関与していることが示唆された。
II)デスミン関連ミオパシー患者の一家系で報告されているαBクリスタリンの変異体であるR120GαBクリスタリンの哺乳動物細胞内における性質について解析した。一過性に発現させたR120GαBクリスタリンは、主に細胞成分の不溶性画分に存在し、細胞内で複数の封入体を形成していること、そして、Hsp27を共発現することでその封入体形成を阻止できることを明らかにした。
III)αBクリスタリンと相互作用する蛋白質を同定するためyeast two-hybrid法によるスクリーニングを開始した。これまでに複数の陽性クローンを得て、それらの全長cDNAを取得し、結合ドメインの解析、組換え蛋白質の精製、培養細胞での発現などを現在行っている。
IV)プロテアソーム阻害剤のラクタシスチンをラット脳室内に連続投与したが、Hsp27は局所的に誘導されてくるが、αBクリスタリンの脳内沈着は観察されなかった。
