Research Abstract

本研究ではVEGF-CおよびVEGF-D産生胃がん細胞株におけるリンパ管増殖誘導能をin vitroおよびin vivoの実験系で検討し、胃がん組織におけるVEGFとその受容体の発現とリンパ管増殖やリンパ節転移との関連性を検討することを目的とする。本年度、胃がん10株を含む21種のヒト培養細胞のVEGF-A、-Cおよび-Dの発現を調べた結果、VEGF-Aは19株、VEGF-Cは5株(胃がんAZ521、TMK-1、線維肉腫HT-1080、前立腺がんPC-3、線維芽細胞KMST-6)に発現が見られたが、VEGF-Dの発現は全く認められなかった。臨床胃がん組織でも約50%にVEGF-Cの発現は認められたが、VEGF-Dの発現は約10%に過ぎず、VEGF-AやVEGF-Cとは異なり、VEGF-Dはがん細胞以外の間質組織または炎症性細胞により産生されることが示唆された。さらに、HT-1080よりVEGF-Aと-C、ヒト肺がんよりVEGF-DのcDNAをクローニングし、作製したVEGF発現ベクターをヒト胃がん細胞(KKLS、MKN-28)に導入した。これらのVEGF過剰発現細胞を用いて受精鶏卵漿尿膜法、ヌードマウス皮下移植および胃漿膜下同所移植系における腫瘍の増殖性をコントロールベクター導入細胞と比較したところ明らかな差は認められなかった。しかし、MKN-28のVEGF-CとVEGF-D発現細胞の皮下腫瘍では血管新生の亢進が認められ、同所移植した腫瘍組織ではリンパ管造成に関与するVEGF受容体の一つであるマウスVEGFR-3遺伝子の発現が増強していた。また、同所移植腫瘍ではヒトVEGF-A遺伝子の発現増強が認められ、VEGF-Cあるいは-Dの過剰発現はパラクリンシステムによりVEGFR-3やVEGF-Aの発現を間接的に亢進することでリンパ管のみならず血管新生も誘導する可能性が示された。
